I'd like to add the area column to the grid-view that is displayed when I use the built-in "My Cases" filter, but the layout change only seems to stick only as long as I don't open another filter.

Is there a way to save changes to the layout of this filter?

No, currently the only way to change this filter would be to modify the source and rebuild FogBugz.

You could save go Filters -> Save Current Filter As... "My Cases, with area". It won't be at the top of the filters list but at least you'll have it saved.
