I've figured out how to do this directly in the database, but was wondering if I am just missing an option in either the User Interface or Plug-in API for setting the homepage of a Wiki.

If you have the Info icon/hyperlink (I believe might be added by the "Wiki Article Info: Details" plugin and located next to the Edit hyperlink on my Wiki) you can "Set Root" under Actions.
